It was programmed so you HAVE to get the Heart Containers after Bosses; otherwise you can't retrieve the pendant/maiden. (The intro level is an exception.)

Also, the glitch that lets you beat the game in 5 minutes does not count; it absolutley requires tool assistance. And tool assistance is a big no-no in the speedrun industry.
