Ad-Aware Personal

Download, install and run. The program will ask for an update. Click OK, then connect. Agree to install, then click Finish when it's done.

You should now be at a flashy-looking, yet somewhat complicated control panel. Just click "Start" and when the next screen shows up, make sure "Smart System Scan" is selected. Click Next.

Now go get a drink and watch some TV, this'll take a while. You'll know it's done when it plays a horrific tone that will frighten your pets. One this is done, click Next.

You'll see a checklist of all the crap Adaware's detected. Right click and click "Select All". Now click Next and enjoy the comfort that comes from Spyware being blasted into space.

This should take care of many problems. However, if you still have issues, try a bunch of programs.

Ad-aware, Spybot, Microsoft Anti-spyware, Hijack-This
Others would probably be fine as well, but remember that some scanners catch things that others don't, and vice-versa, and some scanners work differently and target different types of infection. Hijack-This, for example, helped me get rid of a nasty IE toolbar that Ad-aware and spybot failed to properly remove, but Hijack-This won't target general spyware.

And some anti-virus sollution. If you're investigating a virus or spyware, I also suggest scanning with 2 different virus scanners (AVG and Trend-micro's free online scanner are probably a good bet). Like spyware, virus scanners don't have identical definition lists, and our school district actually had a bad virus outbreak because Symantec (big name virus protection here) didn't have a definition for a virus that Trend Micro was able to identify for us. (And a week later, vice-versa was true with another virus).
Hope this helps. Mods feel free to add onto this topic.

Oh, and if you got the nasties from Kazaa, all I have to say is "lol".

Quick Warning Thanks to Draco:
QUOTE (Draco Icebane)
There are two variants of Ad-Aware that differ in their spelling, one of which is spyware itself
So don't download adaware from anywhere that isn't Lavasoft and you should be safe.

Thanks to Suigi, here's another good program:

I'm kinda peeved that you left out THE best tool for spyware removal: Spybot Search and Destroy!
http://www.safer-networking.org/en/home/index.html
AdAware deals more with adware, Spybot deals more with spyware.
Having both is best.
